研究方向 长期致力于能量代谢稳态调控机制研究,聚焦肥胖、糖尿病、心血管疾病及衰老相关代谢紊乱的分子机制,取得以下原创性成果:(1)首次建立人类遗传性肥胖症的人源干细胞模型,并系统阐明了一系列遗传性肥胖症的致病机理(JCI 2015,2021; JCI Insight 2019; Stem Cell reports 2017);(2)系统鉴定了参与肝糖代谢调控的转录因子,进而发现了新的2型糖尿病 (TOX4) 和非酒精性脂肪肝炎 (PU.1) 的致病因子(Cell metabolism 2022; Journal of Hepatology 2020; Diabetes 2019); (3)揭示IgG作为肥胖和衰老的共同驱动因子,通过介导脂肪组织胰岛素抵抗、慢性炎症及代谢功能失调加速病理进程(Cell Metabolism 2024, 2025)。先后在Cell, Cell Metabolism, Journal of Hepatology,JCI等期刊发表论文20余篇,总被引2400余次。研究成果获JCI同期专题评述,被Nature Aging、Nature Reviews Endocrinology列为研究亮点,其中IgG调控代谢衰老的突破性发现入选Cell Metabolism "解码肥胖复杂系统" 特刊(2024年世界肥胖日纪念专刊),并荣膺"2024年度中国肥胖研究十大成果"。 目前实验室以TOX4和IgG为切入点,整合单细胞转录组学、空间代谢组学、高通量功能基因组学及人诱导多能干细胞(iPSC)模型,重点研究:①能量代谢稳态的神经调控生理及病生理机制;②代谢稳态失衡在衰老及肥胖相关疾病中的病理作用机制;③外周组织糖脂代谢时空特异性调控网络。相关研究为代谢性疾病的精准干预提供新靶点。
